Overview

Melanoma-associated antigen gp100, also known as glycoprotein 100 (gp100), is a melanocyte lineage-specific transmembrane glycoprotein with a molecular weight of approximately 100 kDa. It is highly expressed in melanocytes and melanoma cells, including both cutaneous and uveal melanoma. The protein plays a central role in melanosome maturation—the organelles responsible for melanin synthesis and transport—and is involved in pigmentation processes. It serves as an immunodominant tumor antigen recognized by cytotoxic T lymphocytes (CTLs). Targeting gp100 has led to peptide-based vaccines, mRNA-electroporated dendritic cell therapies, and bispecific fusion proteins like tebentafusp.
