Overview

Melanoma-associated antigen recognized by T cells 1 (MART-1), also known as Melan-A, is a lineage-specific differentiation antigen expressed in melanocytes and the majority of melanoma cases (UniProt Q16655). The MART-1 epitope presented by HLA-A*0201 refers to a specific peptide fragment, such as the natural AAGIGILTV or the optimized heteroclitic analog ELAGIGILTV, bound to the Human Leukocyte Antigen (HLA)-A*0201 molecule (Kawakami et al., 1994, PNAS; Rivoltini et al., 1995, J Immunol). This peptide-MHC complex is a prominent target for cancer immunotherapies, including T-cell receptor (TCR) engineered T-cell therapies and peptide-based vaccines, designed to harness the immune system to eliminate melanoma cells (Johnson et al., 2009, Blood). Because MART-1 expression is largely restricted to melanocytic lineages, it provides a relatively specific target for directing cytotoxic T-cell activity. However, therapeutic targeting of this complex can result in "on-target, off-tumor" toxicities, where the immune system attacks healthy melanocytes in the skin, eyes, and inner ear, leading to conditions like vitiligo, uveitis, and hearing loss (Chhabra, 2011, Immunotherapy).

Mechanism of action

T-cell receptor (TCR) mediated recognition of the peptide-MHC complex on the surface of target cells, leading to T-cell activation, secretion of cytotoxic cytokines (e.g., IFN-gamma, TNF-alpha), and direct cell lysis via perforin and granzymes.
