Overview

Melanoma-associated antigens (MAAs) and Keyhole Limpet Hemocyanin (KLH) represent a combined therapeutic approach used in the development of cancer vaccines for malignant melanoma (National Cancer Institute, 2023). MAAs, including proteins like MAGE-A3 and gp100 or glycolipids like gangliosides GM2 and GD2, are molecules expressed by tumor cells that can be recognized by the immune system (Livingston et al., 1994). However, these antigens are frequently self-antigens and fail to elicit a strong immune response; therefore, they are conjugated to KLH, a highly immunogenic respiratory protein from the giant keyhole limpet (Ragupathi et al., 1996). KLH functions as a carrier and adjuvant, providing potent T-cell help that triggers the production of high-titer antibodies and the maturation of cytotoxic T-lymphocytes specific to the melanoma antigens (Kim et al., 2001). This combination is intended to break immunological tolerance, allowing the patient's immune system to identify and destroy residual cancer cells, thereby reducing the risk of relapse in patients with advanced melanoma (Hoon et al., 1995).

Mechanism of action

KLH acts as an immunogenic carrier and adjuvant that enhances the immune system's recognition of melanoma-associated antigens, leading to the activation of tumor-specific T-lymphocytes and B-lymphocytes (Kim et al., 2001).
