Overview

The melanoma cell membrane is the complex lipid bilayer that defines the boundary of melanoma cells and serves as the primary interface for interactions with the tumor microenvironment (NIH). It is characterized by a unique molecular landscape, including an altered lipidome—such as specific cholesterol and phospholipid ratios—and the overexpression of various melanoma-associated antigens and checkpoint proteins like PD-L1 (Frontiers in Oncology, MDPI). These distinct features make the membrane a target for novel therapeutic strategies, such as membrane-active peptides (e.g., tachyplesin analogues) that selectively bind and disrupt the bilayer to induce cell death (BioRxiv, ResearchGate). Furthermore, the membrane is utilized in biomimetic nanotechnology, where isolated melanoma cell membranes are used to coat drug-loaded nanoparticles to achieve 'homotypic targeting,' allowing the particles to selectively accumulate in tumor tissues by mimicking the surface properties of the cancer cells themselves (MDPI, NIH). In the context of immunotherapy, melanoma cell membranes or lysates have been investigated as vaccine components to stimulate a broad immune response against multiple tumor antigens simultaneously (MDPI). However, because it is a multi-component cellular structure rather than a single molecular entity, it is generally considered a therapeutic platform or a site of multiple targets rather than a discrete pharmacological target. This complexity presents challenges for drug development, particularly regarding off-target toxicity to healthy cell membranes and the potential for systemic side effects like hemolysis (PubMed).
