Overview

MPP7 is a membrane-associated guanylate kinase (MAGUK) scaffold protein in the p55 Stardust subfamily, crucial for the establishment and maintenance of epithelial cell polarity and tight junctions[1][2]. It functions as an adaptor, forming complexes with other polarity proteins such as DLG1 (discs large homolog 1), and indirectly linking to Crb3 via PALS1 (MPP5). Loss of MPP7 disrupts tight junction assembly and epithelial integrity but does not directly result in disease; rather, polymorphisms in the gene are associated with conditions such as epilepsy and autism spectrum disorder[2]. MPP7 is not a classic therapeutic target, enzyme, receptor, or transporter; it lacks known direct interactions with drugs and is primarily implicated in basic cellular organization rather than direct modulation by pharmaceuticals[2][1].
