Overview

Membrane-bound enzymes are a diverse group of catalytic proteins that are physically associated with biological membranes, either as integral proteins spanning the lipid bilayer or as peripheral proteins attached to the membrane surface (NCBI Bookshelf, 2002). These enzymes play fundamental roles in cellular physiology, including the regulation of signal transduction pathways, lipid metabolism, and the processing of extracellular signals (UniProt, 2023). Due to their localization, they are often involved in the first steps of metabolic cascades or the termination of signaling molecules, such as the degradation of neurotransmitters or the activation of peptide hormones. While many specific membrane-bound enzymes, such as Angiotensin-converting enzyme (ACE) or HMG-CoA reductase, are validated therapeutic targets for cardiovascular and metabolic diseases, the term "Membrane-bound enzyme" itself represents a broad functional class rather than a single molecular target (PubMed, 2021). Consequently, pharmacological intervention requires the identification of specific enzyme isoforms to ensure therapeutic precision and avoid widespread off-target toxicity associated with general enzyme inhibition.
