The matrix protein (M) of SARS-CoV-2 is an integral membrane glycoprotein and one of the four major structural proteins. It is the most abundant protein in the viral envelope and plays a central role in virion assembly and morphogenesis. It consists of 222 amino acids with three transmembrane domains, a short N-terminal domain exposed on the exterior, and a long C-terminal domain facing the interior. The M protein forms homodimers and interacts with other viral structural proteins (E, S, N) to direct viral budding. As the most abundant envelope component, M elicits immune responses and serves as a target for antibodies, making it relevant for vaccine design and serological diagnostics.
