Membrane protein BRI3 (BRI3) is a highly conserved, proline-rich transmembrane protein that contains three pairs of cysteine residues within a 42-amino-acid region[4][5]. Its expression is upregulated by Wnt/β-catenin signaling and is thought to play a role in the regulation of cell fate, especially through participation in tumor necrosis factor-alpha (TNF)-induced cell death pathways[1][2][8]. BRI3 has been identified in plasma membrane and azurophil granule membranes and has identical protein binding activity. Experimental evidence links BRI3 to pathways involved in apoptosis and suggests it may modulate both developmental and disease-related cellular processes by integrating Wnt/β-catenin transcriptional responses with apoptotic signaling[1][2][8]. There is limited but emerging evidence of BRI3 involvement in neurodegenerative conditions and cancer; however, it is not currently established as a therapeutic drug target or biomarker[1][2].
