CD20 is a non-glycosylated transmembrane protein expressed on B lymphocytes from the pre-B cell stage through mature stages, but not on early pro-B cells or terminally differentiated plasma cells. It plays a role in calcium flux and B-cell receptor signaling and is a major therapeutic target for monoclonal antibody therapies in hematological malignancies and autoimmune diseases.
Antibody binding induces clustering/oligomerization that recruits complement proteins leading to targeted lysis of B-cells via complement-dependent cytotoxicity.
