Overview

Membrane-spanning 4-domains subfamily A member 15 (MS4A15) is a tetraspanin transmembrane protein localized to the endoplasmic reticulum. It regulates cellular susceptibility to ferroptosis by depleting ER luminal calcium stores and reprogramming lipid metabolism, increasing the abundance of monounsaturated and ether phospholipids that resist oxidative damage. This mechanism provides protection from ferroptosis and supports metabolic adaptation under nutrient stress. MS4A15 is overexpressed in several cancers (notably ovarian cancer), where its high expression is correlated with increased tumor cell proliferation, metabolic reprogramming, and poorer prognosis. Its central role in ferroptosis resistance and metabolic adaptation makes it a promising emerging therapeutic target in cancer biology[1][3][4][7].
