Overview

The membrane-spanning 4-domains subfamily A member 7 (MS4A7) protein is a transmembrane protein of the MS4A gene family, characterized by four membrane-spanning domains and expression in hematopoietic and nonlymphoid cells[1][2][3]. MS4A7’s function appears integral to mature monocyte/macrophage activity and it is implicated as a receptor complex component in signal transduction. Recent studies highlight its pathogenic role in metabolic liver diseases such as MASH, where it is upregulated in TREM2+ macrophages and drives inflammatory injury through activation of the NLRP3 inflammasome. Expression and alternatively spliced isoforms of MS4A7 modulate tissue-specific microenvironments, including tumor and diseased liver contexts. MS4A7 is also associated with Alzheimer’s disease and participates in immune and inflammatory signaling pathways. Ongoing research is exploring MS4A7 as a therapeutic target and biomarker in oncology, neurodegeneration, and metabolic/inflammatory disease settings[1][2][3][4].
