Overview

MenG is an enzyme involved in the biosynthetic pathway of menaquinone (vitamin K2), a key molecule for electron transport in many bacteria. Specifically, MenG catalyzes the final step in menaquinone biosynthesis: the methylation of demethylmenaquinone (DMK) to produce menaquinone (MK). This reaction is essential for generating functional MK, which acts as a major electron carrier during anaerobic respiration. As the menaquinone biosynthesis pathway does not exist in humans, MenG is an attractive antibacterial drug target. Inhibitors targeting MenG have shown promising activity against pathogens like Mycobacterium tuberculosis and can enhance efficacy when combined with existing antibiotics such as rifampicin.
