Overview

Meningococcal Y polysaccharide is the capsular component of Neisseria meningitidis serogroup Y, an encapsulated Gram-negative bacterium responsible for invasive meningococcal disease, including meningitis and septicemia [6, 16]. Structurally, it is a heteropolymer composed of repeating disaccharide units of glucose and N-acetylneuraminic acid (sialic acid) [15]. This polysaccharide is a critical virulence factor that protects the pathogen from host immune defenses by preventing phagocytosis and inhibiting complement-mediated lysis [10]. In medicine, it serves as the primary antigen for various vaccines, including quadrivalent (MenACWY) and pentavalent formulations [5, 8]. Modern conjugate vaccines chemically link this polysaccharide to carrier proteins to induce T-cell dependent immune responses, leading to long-term immunological memory and high-avidity bactericidal antibodies [12, 14]. These vaccines are highly effective at preventing disease and reducing nasopharyngeal carriage in human populations [12].

Mechanism of action

Induction of active immunity by stimulating the production of serogroup-specific bactericidal antibodies (Serum Bactericidal Activity) that facilitate complement-mediated lysis and opsonophagocytosis of Neisseria meningitidis [3, 12].
