Overview

The Mesenchymal stem cell–Biphasic calcium phosphate (MSC-BCP) scaffold interface is a complex biological and material boundary essential for bone tissue engineering (Lobo et al., 2016, PubMed). Biphasic calcium phosphate is a bioceramic composed of hydroxyapatite (HA) and beta-tricalcium phosphate (beta-TCP), designed to mimic the mineral composition of natural bone (Detsch et al., 2015, PubMed). At this interface, MSCs adhere to the scaffold through the adsorption of extracellular matrix proteins like fibronectin, which then bind to cellular integrins (Samavedi et al., 2013, PubMed). This interaction triggers mechanotransduction and biochemical signaling pathways, such as the RUNX2 pathway, leading to osteogenic differentiation and new bone formation. While not a single molecular target, the interface serves as a platform for delivering osteoinductive agents like Bone Morphogenetic Protein 2 (BMP-2) to accelerate healing. The success of this interface is determined by the scaffold's porosity, surface topography, and the rate of ion release, which must be balanced to avoid adverse inflammatory responses or fibrous encapsulation.
