Mesoderm induction early response protein 1 (MIER1) is a transcriptional regulator originally identified as an immediate early gene activated by fibroblast growth factor in mesoderm induction in Xenopus, and is conserved in humans[1][2]. MIER1 functions mainly as a transcriptional repressor by recruiting chromatin-modifying enzymes such as histone deacetylases (HDAC1 and HDAC2), and through interactions with key transcription factors including Sp1 and estrogen receptor alpha (ERα), resulting in repression of their target genes[1][4][2]. MIER1 exists as several alternatively spliced isoforms with differential subcellular localization—important for its activity and linked to development and cancer progression, such as in breast carcinoma where nuclear-cytoplasmic shift of MIER1α is a marker of tumor invasiveness[2]. While MIER1 plays significant roles in gene regulation, chromatin remodeling, development, and cancer, it is not regarded as a classical drug target or therapeutic receptor/enzyme[1][2][5].
