Overview

Mesoderm induction early response protein 3 (MIER3) is a member of the MIER family of transcriptional regulators, containing conserved domains typical of transcription factors such as ELM2 and SANT, and it is predicted to function primarily in the nucleus[1][2]. MIER3 is implicated in the negative regulation of tumor progression—particularly in colorectal cancer and non-small cell lung cancer—by inhibiting cell proliferation, migration, and invasion, promoting apoptosis, and repressing epithelial-mesenchymal transition. Mechanistically, MIER3 acts by directly reducing the activity of the transcription factor Sp1 (in CRC) and the histone acetyltransferase p300, leading to changes in gene expression relevant to tumor suppression. Its down-regulation is associated with poor prognosis and enhanced tumor aggressiveness. While MIER3 is not an established therapeutic target, its loss or reduced expression may serve as a useful prognostic biomarker in certain cancers[1][2].
