Overview

The insulin signaling pathway is a critical biochemical network that regulates glucose and lipid metabolism in response to insulin (Source: StatPearls, Insulin Physiology). The process begins when insulin binds to the Insulin Receptor (INSR), a transmembrane receptor tyrosine kinase, leading to its autophosphorylation and the recruitment of Insulin Receptor Substrate (IRS) proteins (Source: UniProt P06213). These proteins activate the Phosphoinositide 3-kinase (PI3K)/AKT pathway, which facilitates glucose uptake via GLUT4 translocation, and the Ras/MAPK pathway, which governs cell growth and gene expression (Source: KEGG Pathway hsa04910). Metabolic enzymes downstream of this signaling, such as glycogen synthase and glucose-6-phosphatase, are regulated through these cascades to maintain systemic glucose homeostasis (Source: NIH, NIDDK).
