Metabolic enzymes requiring vitamin cofactors are a diverse group of catalytic proteins that necessitate non-protein organic molecules, primarily derived from B-complex vitamins, Vitamin C, and Vitamin K, to perform their biological functions. These enzymes are integral to central metabolic pathways, including the citric acid cycle, nucleic acid synthesis, and the modification of clotting factors (StatPearls, 2023). For example, the enzyme dihydrofolate reductase requires a folate-derived cofactor to facilitate one-carbon transfers necessary for DNA replication, making it a primary target for antifolate chemotherapy (NIH, 2022). Similarly, the gamma-glutamyl carboxylase enzyme requires Vitamin K to activate proteins involved in blood coagulation, a process inhibited by the anticoagulant drug warfarin (PubChem, 2024). Because these enzymes are essential for cellular viability, drugs targeting them often exhibit significant therapeutic efficacy but also carry risks of systemic toxicity and secondary nutritional deficiencies.
