Overview

Metabolic signatures are distinct patterns of metabolites, including small molecules like amino acids, lipids, and carbohydrates, that reflect the physiological or pathological state of a biological system (Nature Reviews Molecular Cell Biology, 2020). These signatures serve as a functional readout of cellular activity, integrating genetic, epigenetic, and environmental influences to provide a snapshot of the current phenotype (Cell Metabolism, 2018). In the context of drug discovery and clinical medicine, they are primarily utilized as biomarkers for disease diagnosis, prognosis, and the assessment of therapeutic efficacy (Journal of Clinical Investigation, 2019). Because they represent a collection of downstream products rather than a single functional entity like a protein or receptor, metabolic signatures are not considered therapeutic targets themselves. Instead, they provide critical insights into the underlying biochemical pathways that may contain druggable enzymes or transporters (NIH, National Human Genome Research Institute). By analyzing these signatures, researchers can identify specific metabolic vulnerabilities in diseases such as cancer or diabetes, guiding the development of precision therapies (Science Translational Medicine, 2021).
