Overview

Metabotropic glutamate receptor 3 (mGluR3) is a member of the group II metabotropic glutamate receptors and is encoded by the GRM3 gene. It is an inhibitory Gi/Go-coupled GPCR localized to presynaptic sites in classical neuronal circuits and, in some cortical regions, postsynaptically. It modulates glutamatergic neurotransmission, synaptic plasticity, and is implicated in learning, memory, anxiety, pain, and neuroprotection. Alterations in mGluR3 signaling are linked to impaired cognition and increased risk for schizophrenia. mGluR3 is a therapeutic target for neuropsychiatric disorders, with ongoing research focusing on selective ligands and allosteric modulators.
