Overview

Metabotropic glutamate receptor 4 (mGluR4) is a G-protein-coupled receptor that modulates neurotransmission by inhibiting adenylyl cyclase. It is primarily expressed in presynaptic terminals in the CNS, particularly in the cerebellum and basal ganglia. Activation or positive modulation of mGluR4 has shown potential therapeutic benefits in Parkinson’s disease, autoimmune disorders, CNS inflammation, and cancer.

Mechanism of action

Upon ligand binding, mGluR4 activates Gi/o proteins, leading to inhibition of adenylyl cyclase and decreased cAMP production, ultimately reducing neurotransmitter release from presynaptic neurons.
