Overview

Metal ion complexes consist of a central metal atom or ion surrounded by molecules or anions known as ligands. In a biological context, these complexes are ubiquitous, forming the functional cores of metalloenzymes, oxygen carriers like hemoglobin, and structural motifs such as zinc fingers. While they are essential for life, the dysregulation of metal ions is linked to various pathologies, including heavy metal poisoning, Wilson's disease, and certain cancers. In pharmacology, metal ion complexes are more frequently categorized as therapeutic agents (e.g., platinum-based chemotherapy) or diagnostic tools (e.g., gadolinium contrast agents) rather than biological targets themselves. When metal ions are targeted, it is typically through chelation therapy to remove excess or toxic metals from the body. Because the term refers to a broad chemical class rather than a specific protein, gene, or pathway, it is generally considered too broad to be a specific therapeutic target in drug discovery.

Mechanism of action

Metal ion complexes typically act as therapeutic agents rather than targets; however, when considered as targets for chelation therapy, drugs bind to free or loosely bound metal ions to facilitate their excretion. In the context of metalloenzymes, drugs may coordinate with the metal center to inhibit catalytic activity.
