Overview

Metallophosphoesterase 1 (MPPE1) is an enzyme essential for the transport and maturation of glycosylphosphatidylinositol (GPI)-anchored proteins. It catalyzes the removal of a side-chain ethanolamine-phosphate (EtNP) from the second mannose of the GPI-anchor protein intermediate, a necessary lipid remodeling step for GPI-anchor maturation. This process is crucial for efficient transport of GPI-anchor proteins from the endoplasmic reticulum to the Golgi apparatus. MPPE1 is primarily localized to the Golgi apparatus and nucleoplasm. While MPPE1 has been associated in genetic contexts with diseases such as Bejel and Yaws, no direct links to cancer, inflammatory, or neurodegenerative diseases are known as of now, nor are any targeting drugs or safety concerns described in current literature[1][3][5].
