Metalloproteases are a broad class of protease enzymes that require a metal ion, most commonly zinc (Zn²⁺), at their active site to catalyze the hydrolysis of peptide bonds in proteins. They play diverse roles across biological systems, including ECM degradation and remodeling, regulation of cell proliferation, migration, differentiation, angiogenesis, apoptosis, and immune responses. Their activities are tightly regulated by endogenous inhibitors known as tissue inhibitors of metalloproteinases (TIMPs). Imbalance between metalloprotease activity and TIMP inhibition can lead to disease.
