Overview

Metastasis-associated protein 1 (MTA1) is a primary component of the Nucleosome Remodeling and Deacetylase (NuRD) complex, functioning as a master regulator of gene expression through chromatin modification (UniProt, Q13330). It acts as a transcriptional coregulator that typically represses tumor suppressor genes, such as the estrogen receptor alpha and p53, thereby promoting oncogenic transformation and progression (PubMed, 12738923). MTA1 is highly expressed in various aggressive malignancies, including breast, prostate, and lung cancers, where it serves as a marker for poor clinical outcomes and high metastatic potential (NCBI, PMC3954012). Its biological roles extend to the regulation of the epithelial-mesenchymal transition (EMT), DNA damage repair, and angiogenesis (PubMed, 24633280). While no FDA-approved drugs specifically target MTA1 alone, it is indirectly modulated by histone deacetylase (HDAC) inhibitors and certain dietary polyphenols like pterostilbene, which are being investigated for their ability to suppress MTA1-mediated signaling (PubMed, 22524488).
