Methionine synthase is an essential enzyme involved in one-carbon metabolism, specifically linking the folate cycle to the methionine cycle. It catalyzes the transfer of a methyl group from 5-methyltetrahydrofolate (5-MeTHF) to homocysteine, resulting in the formation of methionine and tetrahydrofolate (THF). Methionine is an essential amino acid required for protein synthesis and as a precursor for S-adenosylmethionine, a universal methyl donor. Deficiency or dysfunction in methionine synthase leads to elevated levels of homocysteine—a risk factor for cardiovascular disease—and impaired DNA synthesis due to reduced THF availability.
