Overview

Methyl-CpG-binding domain protein 3-like 1 (MBD3L1) is a protein-coding gene product homologous to MBD2 and MBD3, but notably lacks the canonical methyl-CpG-binding domain that enables binding to methylated DNA. Instead, MBD3L1 interacts with members of the NuRD (nucleosome remodeling and deacetylase) complex, specifically MBD2 and MBD3, acting as a transcriptional repressor within certain chromatin contexts. It is expressed predominantly in round spermatids of the testis, suggesting a specialized role in postmeiotic germ cell development. Though it can modulate key gene silencing complexes, knockout studies in mice suggest functional redundancy or a nonessential role under normal conditions. In cell models of facioscapulohumeral muscular dystrophy, DUX4-driven MBD3L1 expression may contribute to loss of required gene repression[1][2][4][6]. There is no evidence supporting MBD3L1 as a clinical therapeutic target or biomarker.
