Overview

Methyl violet is a family of related triarylmethane dyes, most notably comprising methyl violet 2B, 6B, and 10B (also known as crystal violet), which are widely used as histological stains and pH indicators [3, 5, 8]. In a medical context, it is commonly referred to as gentian violet and has been utilized for over a century as a topical antiseptic and antifungal agent for treating various skin and mucous membrane infections [1, 4, 8]. Unlike typical therapeutic targets such as receptors or enzymes, methyl violet is a chemical compound that serves as the active drug or agent itself [3, 6]. Its biological action stems from its ability to penetrate cell membranes and bind to negatively charged molecules like DNA, causing disruption of cellular processes and mitotic inhibition [4, 5]. Recent research has further revealed its potential to inhibit specific proteins such as focal adhesion kinase (FAK) and NADPH oxidase, suggesting possible applications in anti-tumor and anti-angiogenic therapies [1, 2]. However, significant concerns regarding its mutagenic and carcinogenic properties have limited its systemic use and led to stricter regulations [3, 5].

Mechanism of action

Methyl violet acts as an antimicrobial agent by penetrating microbial cell walls and binding to intracellular components like DNA and proteins, which interferes with replication and cellular metabolism [4, 5]. It also dissipates bacterial and mitochondrial membrane potential and has been found to inhibit specific signaling proteins such as Focal Adhesion Kinase (FAK) and enzymes like NADPH oxidase [1, 2, 4].
