Methylenetetrahydrofolate dehydrogenase 1-like (MTHFD1L) is a mitochondrial monofunctional enzyme possessing formyl-tetrahydrofolate synthetase activity critical to the folate cycle and one-carbon metabolism. It generates formate in mitochondria, enabling nucleotide and methionine biosynthesis, and is implicated in the growth and progression of multiple cancers through its role in cellular metabolism. Overexpression of MTHFD1L is associated with poor prognosis in several tumor types and affects immune cell infiltration in the microenvironment.
