Methylenetetrahydrofolate dehydrogenase 1-like (MTHFD1L) is a mitochondrial monofunctional enzyme possessing formyl-tetrahydrofolate synthetase activity critical to the folate cycle and one-carbon metabolism. It generates formate in mitochondria, enabling nucleotide and methionine biosynthesis, and is implicated in the growth and progression of multiple cancers through its role in cellular metabolism.
