Methylthioribulose-1-phosphate dehydratase (APIP) is an enzyme encoded by the APIP gene in humans, primarily known for its role in the methionine salvage pathway, catalyzing the dehydration of methylthioribulose-1-phosphate to 2,3-diketo-5-methylthiopentyl-1-phosphate. APIP also functions in the regulation of apoptosis, as it interacts with APAF1 and can inhibit both caspase-1-dependent inflammatory cell death (pyroptosis) and caspase-9-dependent apoptosis, suggesting a potential role in controlling cell fate under stress and disease conditions. APIP deficiency is associated with increased cell death and has been linked to cancer pathogenesis and other diseases related to aberrant cell survival or death. The protein is localized in the cytoplasm and is widely expressed in tissues.
