Overview

MF59 is a licensed, squalene-based oil-in-water emulsion vaccine adjuvant used to enhance the immunogenicity of co-administered antigens, particularly in influenza vaccines. It does not directly interact with a single molecular target (such as a receptor or enzyme), but rather functions by inducing local and lymph node innate immune activation. This involves rapid recruitment of innate immune cells, induction of pro-inflammatory chemokines and cytokines, and the creation of an "immunocompetent" environment that leads to better antigen presentation and stronger adaptive (antibody and T cell) responses. MF59’s activity is partly mediated by the release of endogenous danger signals (such as ATP) from damaged muscle cells, rather than direct pattern recognition receptor (PRR) engagement. It is approved for human use and has a strong safety record in both elderly and pediatric populations[1][2][3][4][5][6][7].
