MG7 tumor-associated antigen (MG7-Ag) is a highly specific glycoprotein marker primarily associated with gastric cancer and its precancerous states. It was originally identified using the MG7 monoclonal antibody, which demonstrates high sensitivity and specificity for malignant gastric cells compared to normal gastric mucosa. Biologically, MG7-Ag is involved in promoting tumor cell proliferation, migration, and invasion, making it a critical factor in the progression of gastrointestinal malignancies. Its expression often correlates with poor prognosis and advanced clinical stages in patients. In the therapeutic landscape, MG7-Ag serves as a versatile target for various modalities, including monoclonal antibodies, immunotoxins, and chimeric antigen receptor (CAR) T-cell therapies. Because it is expressed early in the transition from dysplasia to carcinoma, it is also utilized as a biomarker for the early screening and diagnosis of gastric cancer. Research into MG7-Ag mimotope vaccines has further highlighted its potential for active immunotherapy, aiming to stimulate the host immune system to recognize and eliminate MG7-positive tumor cells while sparing healthy tissue.
Targeting of MG7-Ag via monoclonal antibodies or CAR-T cells induces antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), or direct T-cell mediated lysis of tumor cells expressing the antigen. Mimotope vaccines aim to induce a specific humoral and cellular immune response against MG7-expressing malignant cells.
