Overview

MIA SH3 domain ER export factor 2 (MIA2) is a membrane-associated, SH3-domain containing protein encoded by the MIA2 gene, primarily functioning as a receptor in the endoplasmic reticulum involved in the export of large cargo such as pre-chylomicrons and pre-very low-density lipoproteins (pre-VLDLs)[4]. MIA2 is part of a gene family that includes transcript variants specific to melanoma inhibitory activity 2, cTAGE5, and TANGO1-like isoforms, which reflects the gene’s complex splicing and multifunctional nature. It acts as a cargo receptor coordinating with other ER exit site proteins to facilitate vesicle-mediated transport of select proteins and lipoproteins from the ER to the Golgi apparatus[4]. Disease roles are implicated in various cancers—especially as a marker in melanoma, meningioma, and lymphoma[4]. While it is designated by multiple aliases (CTAGE5, TALI, MEA6, among others), its key biological functions are centered on protein trafficking within secretory pathways. There are currently no approved therapeutics specifically targeting MIA2, and its mechanism of action in pathologies remains an active area of research[4].
