MICOS complex subunit MIC27 (APOOL) is a mitochondrial inner membrane protein, localized to the mitochondrial intermembrane space, and functions as an integral component of the MICOS (Mitofilin/MINOS) complex[1][2][3][5]. APOOL specifically binds the mitochondrial lipid cardiolipin, and this interaction is important for the structural maintenance of mitochondrial cristae—membrane foldings critical for mitochondrial respiration[1][2][3]. APOOL physically interacts with other essential subunits of the MICOS complex, including Mitofilin, MINOS1, and SAMM50, influencing the shape and function of cristae[2][3][5]. Overexpression or downregulation of APOOL disrupts mitochondrial architecture and respiration, highlighting its role in organelle homeostasis and inner mitochondrial membrane organization[1][2][3]. There is currently no strong evidence that APOOL is a direct therapeutic target, nor are there clinically relevant drugs or biomarker strategies targeting this protein in disease, though its homolog APOO has been implicated in cholesterol metabolism and cardiovascular disease in model organisms[4].
