Overview

Microbial and mucosal cell membranes are complex lipid bilayers that serve as the fundamental structural boundaries for both pathogenic organisms and host epithelial cells. In pharmacology, the microbial membrane is a critical target for several classes of antimicrobial agents, such as polymyxins, lipopeptides, and polyenes, which exploit differences in lipid composition to achieve selective toxicity (NIH, 2023). These drugs typically interact with specific molecules like lipopolysaccharides in Gram-negative bacteria or ergosterol in fungi to induce membrane permeabilization, pore formation, and rapid cell death (PubChem). However, the mucosal aspect refers to the host's own cell membranes, which can be inadvertently damaged by these agents due to structural similarities, leading to significant side effects such as nephrotoxicity or local tissue irritation (StatPearls, 2024). Consequently, therapeutic development in this area focuses on optimizing the therapeutic index by maximizing affinity for microbial-specific lipids while minimizing interactions with the neutral phospholipids characteristic of human mucosal surfaces (PubMed).
