Microbial and tissue components refer to a broad category of biological molecules that serve as triggers for the innate immune system. This group includes Pathogen-Associated Molecular Patterns (PAMPs), such as lipopolysaccharides and viral nucleic acids, as well as Damage-Associated Molecular Patterns (DAMPs), such as HMGB1 and heat shock proteins released during cellular stress [Janeway & Medzhitov, 2002; Kumar, 2020]. These components are recognized by Pattern Recognition Receptors (PRRs), including Toll-like receptors (TLRs), which subsequently activate inflammatory pathways and shape the adaptive immune response [Akira et al., 2006]. In clinical pharmacology, these components are utilized as active ingredients in vaccines and immunomodulators, such as the BCG vaccine for bladder cancer or bacterial lysates for respiratory health, to enhance immune surveillance [StatPearls, 2023]. Conversely, certain drugs like Polymyxin B are designed to bind and neutralize specific microbial components like endotoxins to treat severe conditions such as sepsis [PubChem]. Because this term describes a heterogeneous class of ligands rather than a specific molecular entity, it is not classified as a canonical therapeutic target.
These components function as ligands for Pattern Recognition Receptors (PRRs), such as Toll-like receptors (TLRs) and NOD-like receptors (NLRs), to initiate innate immune signaling or are neutralized by drugs to prevent excessive inflammatory responses [Akira et al., 2006; Kumar, 2020].
