Overview

The microbial cell membrane and cell wall interface is a critical structural and functional zone in bacteria where the cytoplasmic membrane meets the peptidoglycan cell wall. This interface is the site of essential biosynthetic processes, most notably the final stages of cell wall assembly involving the translocation and polymerization of peptidoglycan precursors like Lipid II. Many potent antibiotics, including glycopeptides (e.g., vancomycin) and lipopeptides (e.g., daptomycin), exert their bactericidal effects by targeting molecules or physical properties at this junction. By disrupting the integrity of the membrane or sequestering precursors necessary for cell wall expansion, these drugs cause rapid cell death and lysis. This target is particularly significant in the treatment of multidrug-resistant Gram-positive infections, as it involves highly conserved components that are absent in human cells, providing a high degree of selective toxicity. The interface also plays a role in cell division and the spatial organization of the divisome and elongasome complexes.
