Microbial cell membranes and structural proteins represent a broad class of essential components required for the survival, integrity, and reproduction of pathogenic microorganisms. The cell membrane serves as a vital semi-permeable barrier and a scaffold for bioenergetic processes, while structural proteins, such as penicillin-binding proteins (PBPs), are integral to the assembly of the protective cell wall (StatPearls, 2023). These components are highly effective therapeutic targets because they often possess unique biochemical properties distinct from human cells, allowing for selective toxicity (NIH, 2022). For example, lipopeptide antibiotics like daptomycin insert into the bacterial membrane to cause depolarization, while beta-lactams target structural PBPs to inhibit cell wall cross-linking (PubMed, 2021). Despite their utility, drugs targeting these structures can face challenges such as nephrotoxicity or the rapid development of microbial resistance through membrane remodeling (NCBI, 2020). Understanding these targets is fundamental to the development of narrow and broad-spectrum anti-infective therapies.
