Overview

Microbial cell membranes and viral envelopes are essential lipid-based structures that define the boundary of pathogens and maintain their physiological integrity (Nature Education, 2014). In bacteria and fungi, the cytoplasmic membrane is responsible for osmotic regulation, nutrient transport, and energy production through the electron transport chain (StatPearls, 2023). Viral envelopes, acquired from host cell membranes during budding, contain viral proteins necessary for host cell recognition and fusion (NCBI, 2000). These structures are prime targets for antimicrobial agents because they often contain unique components, such as ergosterol in fungi or lipopolysaccharides in Gram-negative bacteria, which are absent in human cells (PubMed, 2017). Therapeutic agents like polymyxins and polyenes exploit these differences to induce membrane leakage or pore formation, resulting in rapid pathogen neutralization (StatPearls, 2023). However, the structural similarity between certain microbial and host lipids can lead to toxicity, such as nephrotoxicity or hemolysis, necessitating careful clinical management (NIH, 2022).
