Microbial cell surface and membrane structures encompass the essential outer layers of bacteria, fungi, and viruses, including cell walls, plasma membranes, and envelopes. These structures provide mechanical strength, regulate the transport of nutrients and waste, and facilitate interactions with the host environment (StatPearls, 2023). In bacteria, the peptidoglycan layer is a primary target for beta-lactam antibiotics like penicillin, which inhibit cross-linking and lead to osmotic lysis (NIH, 2022). Fungal membranes are characterized by the presence of ergosterol, a sterol targeted by polyene and azole antifungals to disrupt membrane fluidity and integrity (PubMed, 2021). Additionally, certain antibiotics like polymyxins act as detergents to destabilize the outer membrane of Gram-negative bacteria. Because these structures often contain components unique to microorganisms, they offer high selectivity for antimicrobial therapy. However, the emergence of resistance through structural modifications remains a significant clinical challenge (Nature Reviews Microbiology, 2020).
