Microbial lipid membranes are essential biological structures that serve as the primary osmotic barrier and the site for vital metabolic processes, including energy production and cell wall synthesis [1]. These membranes differ significantly from mammalian membranes in their lipid composition; while mammalian membranes are rich in neutral lipids and cholesterol, microbial membranes often contain high proportions of negatively charged lipids like phosphatidylglycerol or specific sterols like ergosterol [2,3]. These biochemical differences provide a basis for selective toxicity, allowing drugs to target microbial cells specifically [4]. For instance, the lipopeptide daptomycin binds to bacterial membranes in a calcium-dependent manner to cause depolarization, while polyene antifungals like amphotericin B bind to ergosterol to form lethal pores [5,6]. Because the membrane is a fundamental structural component, it remains a critical target for overcoming multidrug-resistant pathogens [7]. However, maintaining high selectivity remains a challenge, as some membrane-active agents can cause significant host toxicity, such as the renal damage observed with polymyxins [8].
Drugs targeting microbial lipid membranes generally act through electrostatic attraction to anionic lipids, followed by membrane insertion, pore formation, and subsequent depolarization or physical disruption of the bilayer integrity [4,5].
