Overview

Microbial lipid membranes and their associated proteins constitute the essential structural and functional boundary of bacteria, fungi, and other microorganisms. These membranes are composed of a lipid bilayer—often containing unique components like lipopolysaccharides (LPS) in Gram-negative bacteria or ergosterol in fungi—and a diverse array of proteins including transporters, enzymes, and receptors [1, 2]. They serve critical roles in maintaining cellular homeostasis, energy production via the electron transport chain, and the synthesis of the cell wall [3]. Because these structures differ significantly from mammalian cell membranes, they are primary targets for various antimicrobial agents [4]. Drugs like polymyxins and daptomycin disrupt membrane integrity by binding to specific lipid components, while others target membrane-associated enzymes like penicillin-binding proteins (PBPs) to inhibit cell wall assembly [5, 6]. This target complex is central to the mechanism of action for many of the most clinically significant antibiotics and antifungals used today.
