Overview

Microbial proteins and lipoproteins are a broad class of molecules derived from bacteria, viruses, fungi, and other pathogens that play critical roles in microbial physiology and host-pathogen interactions (NIH). They are primarily recognized by the host's innate immune system as Pathogen-Associated Molecular Patterns (PAMPs), with lipoproteins being the classic ligands for Toll-like receptor 2 (TLR2) heterodimers (StatPearls). In clinical practice, specific microbial proteins are the targets of various antimicrobial agents, such as beta-lactams and macrolides, which disrupt essential bacterial functions like cell wall synthesis and translation (DrugBank). Additionally, these molecules serve as the antigenic basis for most vaccines and are the targets of neutralizing monoclonal antibodies used to treat infectious diseases (PubMed). While the term encompasses a vast array of distinct molecules, they collectively represent the primary interface through which the host detects and responds to microbial threats.

Mechanism of action

Microbial proteins and lipoproteins act as ligands for host Pattern Recognition Receptors (PRRs), such as TLR2, to initiate innate immune responses. Specific microbial proteins are targeted by antibiotics to inhibit essential cellular processes or neutralized by antibodies to prevent infection.
