Overview

Microbial structural components and toxins encompass a diverse group of molecules that are fundamental to the viability and pathogenicity of infectious agents. Structural components, such as the peptidoglycan layer in bacteria and the ergosterol in fungal membranes, provide mechanical strength and are essential for survival, making them ideal targets for antibiotics like penicillins and antifungals like amphotericin B (StatPearls, 2023). Toxins, including both endotoxins (lipopolysaccharides) and exotoxins (secreted proteins), are primary drivers of disease symptoms by damaging host cells or inducing excessive immune responses (Nature Reviews Microbiology, 2016). Therapeutic strategies often involve the direct neutralization of these toxins using monoclonal antibodies, such as bezlotoxumab for Clostridioides difficile or raxibacumab for anthrax (NEJM, 2017). While targeting these components is highly effective for treating infections, it also presents challenges such as the development of antimicrobial resistance and the risk of systemic inflammation upon the rapid lysis of pathogens. Consequently, these targets remain central to the development of both traditional anti-infectives and modern biologics aimed at mitigating virulence.
