Overview

Microfibril-associated glycoprotein 2 (MAGP-2), encoded by the MFAP2 gene, is a 25 kDa extracellular matrix protein found in microfibrils and is involved in their assembly and function[1][3]. MAGP-2 binds to fibrillin-1 and fibrillin-2, modulates extracellular matrix structure, and plays a role in regulating availability of growth factors such as TGFβ and BMPs[1][4]. Unlike MAGP-1, MAGP-2 contains an RGD integrin-binding motif, enabling interactions with αvβ3 integrins[1]. Its tissue expression is more restricted than MAGP-1. Disease associations include roles as a biomarker and contributor to cancer invasiveness, immune response regulation, and metabolic and cardiovascular disorders, primarily elucidated in animal models and correlative studies in human cancers[5][10][11]. There are no known approved drugs targeting MAGP-2 or clinical trials directly modulating its activity.
