Overview

Microfibrillar-associated protein 5 (MFAP5) is a 25-kDa extracellular matrix glycoprotein, predominantly involved in the structure and function of microfibrils that support elastic fibers in connective tissues[5][6][7]. It promotes attachment of cells to microfibrils via alpha-V-beta-3 integrin, participates in elastinogenesis, and is critical for vessel wall integrity and regulation of growth factor signaling in the cardiovascular system[2][7]. MFAP5 also plays roles in hematopoiesis and has been implicated in the regulation of adipogenesis by inhibiting coactivators of PPARγ[1][4]. Dysregulation or mutation of MFAP5 is associated with diseases such as familial thoracic aortic aneurysm, several cancers, obesity-related tissue remodeling, and neutropenia[1][3][6][7]. It is an emerging therapeutic target due to its functions in disease-related extracellular matrix dynamics and cell-matrix interactions[2][3][6].
