Overview

Microglial cells are the resident immune cells of the central nervous system (CNS), acting as the first line of defense against pathogens and brain injury (Source: NIH, StatPearls). Originating from yolk sac-derived myeloid progenitors, they maintain CNS homeostasis by performing immune surveillance, debris phagocytosis, and synaptic pruning during neurodevelopment (Source: PubMed). In the context of pathology, microglia can transition between various functional states; while acutely protective, chronic microglial activation is a hallmark of neurodegenerative diseases like Alzheimer's and Parkinson's, where they drive neuroinflammation and contribute to neuronal loss (Source: Nature Reviews Neurology). Although technically a cell type rather than a single molecular target, they express various receptors and enzymes—such as TREM2, CSF1R, and TSPO—that are the actual focus of modern drug discovery efforts (Source: Science Translational Medicine).
