Microglial targets encompass a diverse array of proteins, including receptors, enzymes, and ion channels, that are expressed by microglia, the primary immune cells of the central nervous system (CNS). These targets, such as TREM2, CSF1R, and P2X7, play critical roles in maintaining CNS homeostasis through functions like phagocytosis, synaptic pruning, and the release of neurotrophic factors (Deczkowska et al., 2018, Cell). In the context of neurodegenerative diseases like Alzheimer's and Parkinson's, microglia often transition into a disease-associated state characterized by chronic neuroinflammation and impaired clearance of toxic protein aggregates (Keren-Shaul et al., 2017, Cell). Pharmacological intervention aimed at these targets seeks to shift microglia from a neurotoxic pro-inflammatory state to a neuroprotective or homeostatic state. For example, CSF1R inhibitors like pexidartinib can deplete or modulate microglial populations, while TREM2-targeting antibodies are being investigated to enhance the clearance of amyloid-beta (Ulland & Colonna, 2018, Nature Reviews Immunology). Other targets like the P2X7 receptor are being explored to block the maturation and release of pro-inflammatory cytokines like IL-1β (Burnstock & Knight, 2018, Frontiers in Pharmacology).
