MicroRNA 101-2 is a highly conserved, small non-coding RNA encoded in the human genome at chromosome 9p24.1, serving as one of two precursor transcripts for mature miR-101. It regulates gene expression post-transcriptionally by binding the 3’UTR of target messenger RNAs, resulting in mRNA degradation or translation inhibition. MiR-101-2 frequently acts as a tumor suppressor, directly repressing multiple oncogenes (e.g., EZH2, COX-2, MCL-1, FOS, USP47, KPNA2) and is involved in critical biological processes such as inhibiting cell proliferation, migration, invasion, and promoting apoptosis. The molecule is notably downregulated in a range of malignancies and other progressive diseases including fibrotic and cardiovascular conditions, making its restoring a potential approach in miRNA-based therapeutics and biomarker development.
Restoration or overexpression of miR-101-2 inhibits oncogenic targets (EZH2, COX-2, MCL-1, FOS, USP47, KPNA2), leading to suppression of tumor proliferation, migration, invasion, and promotion of apoptosis. miR-101-2 mimics act as gene therapy agents to modulate downstream gene expression.
