Overview

MicroRNA 10b-5p (miR-10b-5p) is a small non-coding RNA that functions as a master regulator of metastatic progression in various human cancers (Ma et al., 2007, Nature). It is processed from the MIR10B gene located within the HOX gene cluster and acts by binding to the 3' untranslated regions (UTRs) of target mRNAs to inhibit their translation or induce degradation. A primary mechanism involves the suppression of Homeobox D10 (HOXD10), which leads to the upregulation of the pro-metastatic protein RHOC, facilitating tumor cell migration and invasion (PubMed, PMID: 17898770). miR-10b-5p is highly overexpressed in metastatic breast cancer, glioblastoma, and pancreatic cancer, making it a valuable diagnostic and prognostic biomarker (NIH, National Cancer Institute). Therapeutic targeting of miR-10b-5p is currently being explored using antagomirs, such as TTX-MC138, which are antisense oligonucleotides designed to sequester the microRNA and restore the expression of its target genes (TransCode Therapeutics, 2023). These therapeutic interventions aim to halt the spread of cancer cells to distant organs, addressing a critical need in oncology.
